Each real-time system differs in its configuration, however they are all similar in so much as they detect emitted fluorescence for each reaction from the specific fluorophore(s) being used. For the purpose of this discussion we will simply be referring to two methods, DNA binding dyes and probes. We are aware that there are subgroups within these categories but they are discussed elsewhere in detail in this book. For detailed guidance on the selection of an appropriate detection method (binding dye or probe) the reader is referred to the relevant chapters of this book.
