To provide easy access to our computationally designed primers, we have constructed an online primer database, PrimerBank with a freely accessible web interface (http://pga.mgh.harvard.edu/primerbank). Figure 5.2 is a screen shot of the primer search page. There are several ways to search for primers: GenBank Accession, NCBI protein accession, gene ID, PrimerBank ID or Keyword. PrimerBank currently contains over 300,000 primers designed for most known human and mouse genes. This web site also presents a detailed experimental protocol for SYBR® Green real-time PCR and common issues concerning real-time PCR. A short version of the protocol is given later in this chapter.
