Sample preparation

1. Cell lines or blood cells - For attached cells, first trypsinize or scrape attached cells from the culture flask. Pellet suspension cells by centrifu-gation. Determine cell number using a hematocytometer and place ~3 x 106 cells into to a labeled, microcentrifuge tube. Centrifuge for 5 min at 600 x G, aspirate the supernatant and store the tubes at -80°C.

2. Tissues - Place 10 mg of tissue (2 mm3) into a microcentrifuge tube. Flash freeze the tube by dropping it into a dewer containing liquid nitrogen and store the tubes at -80°C.
